Imaging Glucose Oxidase by Scanning Electrochemical Microscopy

چکیده

Scanning Electrochemical Microscopy is a tool for imaging and investigate surfaces by meaning of chemical reactivity, electric conductivity, and enzymatic activity. This technique is typical electrochemical cell plus three axis positioner. Positioner allows to measure typical electrochemical measurements, like Cyclic Voltammetry, Chronoamperometry, Impedance Spectroscopy, etc. in the vicinity of substrate and in the chosen position of working electrode. Moreover, this technique can be used as biosensor, e.g. measuring glucose concentration or other parameters, such as lactate and oxygen detection in living and unhealthy cells. In this paper, we present imaging of glucose oxidase by Scanning Electrochemical Microscopy in feedback mode. For experiments, glucose oxidase on the plastic surface was used as well-known enzyme. Imaging of glucose oxidase was performed in buffer with different glucose concentrations and with different redox mediator concentrations. Measurements showed that for higher image resolution glucose concentration has to be about 15-20 mmol·L -1 and redox mediator concentration needs to be lower than 0.2 mmol·L -1 . For image processing, Matlab programme was used. Another part of measurements was approaching the ultramicroelectrode to the surface and the current dependence on distance is shown in different concentrations of redox mediator and glucose. Scanning electrochemical microscopy. 24. Enzyme ultramicroelectrodes for the measurement of hydrogen peroxide at surfaces.

Hydrogen peroxide sensing microelectrodes for scanning electrochemical microscopy (SECM) have been developed and used to measure hydrogen peroxide in the diffusion layer during electrochemical reduction of oxygen on gold and carbon electrodes. These microelectrode biosensors were also used to detect immobilized glucose oxidase through the production of hydrogen peroxide during enzymatic oxidati...

Scanning electrochemical microscopy. 23. Reaction localization of artificially patterned and tissue-bound enzymes.

The scanning electrochemical microscope (SECM), operating in the feedback mode, was used to image localized surface reactions of redox enzymes at the micrometer level. Surfaces imaged with the SECM included glucose oxidase immobilized within 8-microns-diameter pores of a filtration membrane and individual whole mitochondria with active NADH cytochrome reductase enzymes in their outer membranes....

Spatially addressed deposition and imaging of biochemically active bead microstructures by scanning electrochemical microscopy.

A new procedure is described to deposit paramagnetic beads on surfaces to form microscopic agglomerates. By using surface-modified beads, microscopic structures with defined biochemical activity are formed. The shape and size of agglomerates were characterized by scanning electron microscopy (SEM), and the biochemical activity was mapped with scanning electrochemical microscopy (SECM). This app...

Imaging of biological macromolecules on mica in humid air by scanning electrochemical microscopy.

Imaging of DNA, keyhole limpet hemocyanin, mouse monoclonal IgG, and glucose oxidase on a mica substrate has been accomplished by scanning electrochemical microscopy with a tungsten tip. The technique requires the use of a high relative humidity to form a thin film of water on the mica surface that allows electrochemical reactions to take place at the tip and produce a faradaic current (approxi...

Fabricating and imaging carbon-fiber immobilized enzyme ultramicroelectrodes with scanning electrochemical microscopy.

The scanning electrochemical microscope (SECM) is used to image the activity of enzymes immobilized on the surfaces of disk-shaped carbon-fiber electrodes. SECM was used to map the concentration of enzymatically produced hydroquinone or hydrogen peroxide at the surface of a 33-microm diameter disk-shaped carbon-fiber electrode modified by an immobilized glucose-oxidase layer. Sub-monolayer cove...
